English version

Conclusion. Seals on sterile barrier packaging are usually made by heat sealing, with temperature, pressure and dwell time as the principal parameters. All three vary in production, so process validation must answer not whether a particular setting works but over what range settings work. That range is the parameter window, and its significance is that so long as production parameters fall within it, seal quality is acceptable. Too narrow a window and ordinary equipment variation takes the process outside it, producing batch defects; a window insufficiently validated may include parameters that are in fact marginal.

Effects of the parameters. Temperature too low gives insufficient fusion and a weak seal, while too high over-melts the material, reducing strength or perforating it. Pressure too low gives poor contact and a discontinuous seal, while too high thins the material and reduces strength. Dwell too short gives insufficient fusion, while too long conducts excessive heat and affects surrounding material. Note that all three fail both low and high; this is often misread as more being safer, when excessive temperature over-melts or perforates and excessive pressure thins the seal. The three also interact: longer dwell partly compensates for lower temperature, and changing pressure alters heat transfer efficiency, so the window cannot be established by varying one parameter alone.

Establishing the window. Define the range to examine, taking a range around the setting wide enough to cover possible variation. Design the test points as a matrix of parameter combinations. Produce sufficient samples at each combination. Test them for seal strength and integrity. Analyse the results to find the region where everything passes, which is the window. And fix the setting and tolerance, choosing a robust point within the window and stating the permitted variation, with the setting as near the centre of the window as possible. Centring the setting is important practice: set at an edge, variation in one direction leaves the window quickly, while set at the centre both directions have margin.

Test methods. Seal quality testing divides into two categories. Strength tests, covering seal strength as peel force and burst strength, show how robust the seal is. Integrity tests, covering dye penetration, bubble testing and leak testing, show whether channels exist. Both are needed: adequate strength does not mean the absence of a fine channel, and a fine channel suffices to admit microorganisms, making integrity the more direct indicator of sterility assurance. Visual inspection of seal uniformity, wrinkles and entrapped matter is the most basic check and, though simple, catches obvious problems.

Other factors affecting the window. Packaging material batch variation in thickness and coating affects sealing. Equipment condition matters through tooling wear, heater ageing and pressure system drift. Environmental temperature and humidity may have an effect. The product itself may affect loading in the seal area through its shape and rigidity. And contamination in the seal area from product residue or dust creates channels. Foreign matter in the seal area is a common production defect: product debris, packaging flash and contamination during handling can all be trapped in the seal and form a channel. Parameter control cannot address this; procedural discipline and inspection must.

When to revalidate. After packaging material changes in material, supplier or specification. After equipment change or overhaul. After product changes affecting the seal area. After a change of sterilisation method, since different methods affect seals differently. And at the scheduled interval. Sterilisation change deserves attention: seal performance before and after sterilisation can differ, since irradiation may embrittle materials and moist heat pressure changes may affect seals. Test seals after sterilisation, or both before and after.

Routine control. Monitor parameters, recording actual values per batch and confirming they lie within the window. Inspect the first unit after start-up or changeover. Inspect during the process, sampling seal strength and integrity at a defined frequency. Check equipment daily, covering tooling, heating and pressure systems. Inspect incoming packaging material. And define the handling of parameters outside the window. First-unit inspection is an effective control point, since equipment behaves differently at start-up than in steady running and the check catches start-up problems.

Relationship to stability. Seal performance changes during storage. Materials age and seal strength may decline. Internal stress in the seal relaxes over time. Storage temperature and humidity have effects. And transport vibration and shock act on the seal. Include seal performance in stability studies, testing strength and integrity at intervals across the shelf life. Data from the point of manufacture do not represent the state at the end of shelf life.

How we handle it. For seal verification we suggest both strength and integrity testing, since they reveal different problems and strength testing alone can miss fine channels. For sterile products we suggest testing seals after sterilisation, or before and after for comparison, since sterilisation genuinely affects seals and pre-sterilisation data do not represent the final product.
