English version

Conclusion. Moist heat sterilisation transfers heat by saturated steam condensing on item surfaces, so its precondition is that steam must reach every surface to be sterilised. That sounds simple and is in fact the root of most failures: air not fully removed, packaging obstructing steam entry, loading so dense that steam cannot reach some positions, and lumens that cannot be displaced. In such cases a temperature probe may indicate that the set value has been reached while sterilising conditions have not in fact been established everywhere. The core of validation is therefore not whether the temperature was reached but whether the steam arrived.

Key parameters. Temperature, examined for value and uniformity across positions. Time, meaning the hold after temperature is reached. Steam quality, covering dryness, superheat and non-condensable gases. Pressure, examined against temperature. Air removal, meaning the effectiveness of the vacuum phase. Loading, covering density, arrangement and packaging. And cooling and drying, covering the cooling process and residual moisture. The pressure-temperature relationship is a useful diagnostic: saturated steam has a one-to-one correspondence between temperature and pressure, and a measured temperature below the saturation temperature for that pressure indicates non-condensable gas, usually air, in the chamber.

Content of validation. Empty chamber temperature distribution establishes uniformity and locates cold spots. Loaded temperature distribution establishes the distribution under actual loading, including within items. Penetration verification establishes whether steam reaches item interiors, particularly inside packaging and lumens. Biological indicator verification confirms sterilising effectiveness using appropriate indicators placed at the more difficult positions. Air removal verification confirms the vacuum phase. Loading validation covers every permitted arrangement. And drying verification measures residual moisture after the cycle. Locating cold spots is the key action: once found, routine monitoring points belong at the cold spot, since conditions met there are met elsewhere.

Common sources of deviation. Incomplete air removal, from declining vacuum pump performance, leaks or pipework problems, which is the most frequent cause. Poor steam quality, either too wet with high water content or superheated above saturation temperature, both impairing heat transfer. Overly dense loading, leaving insufficient gaps for steam circulation. Inconsistent loading, arranged differently each time so that results are not reproducible. Inappropriate packaging, either insufficiently permeable or wrapped too tightly. Lumens and blind holes, where air is trapped and cannot escape. Poorly placed probes, positioned where temperature is easily reached so that the true cold spot is not measured. And condensate accumulation, where the arrangement lets water collect and impairs drying. Inconsistent loading is the actual situation in many companies: validation is performed one way while operators arrange loads their own way in production, and the validation conclusion then does not apply to production. Fix the loading arrangement with illustrated instructions specifying placement, quantity and position, and enforce it in production.

Routine monitoring. Record and retain temperature, pressure and time traces for every batch. Use chemical indicators in every batch at specified positions. Use biological indicators at the specified frequency. Perform air removal tests periodically. Maintain equipment, including vacuum pumps, seals, steam traps and filters. Monitor steam quality periodically. And check that loading matches the validated arrangement. Maintenance strongly affects stability: pump performance declines gradually and seals age gradually, and neither causes immediate failure while both erode the safety margin until some batch fails.

When to revalidate. After major overhaul or replacement of key components. After a change of loading arrangement. After a change of packaging material or method. After a change to product structure or materials. After parameter adjustment. And at the scheduled revalidation interval. Packaging changes are easily overlooked: a different packaging material may have different permeability and steam penetration changes accordingly, and producing without revalidating carries real risk.

Product compatibility. The process must suit the product. Material tolerance determines whether the product withstands the temperature and humidity. Structural design should avoid blind holes and closed cavities that cannot vent. Packaging design balances permeability against microbial barrier. Arrangement convenience depends on whether the product's shape allows consistent placement. And drying convenience depends on whether the structure traps water. These cost little to address during product design and may require tooling and process changes afterwards.

How we handle it. For moist heat work we attend to two things: whether probes are genuinely placed at the difficult positions, and whether the pressure-temperature relationship is normal. The first determines whether the data are representative and the second points directly to non-condensable gas problems. For material tolerance verification we suggest processing at the upper limit of permitted parameters and re-measuring mechanical performance and function afterwards, since moist heat effects on polymers can be cumulative.
